In a conceptually current, quick-reference, Question & Answer format, the Handbook of Practical Immunohistochemistry: Frequently Asked Questions provides standardization of the immunostaining process for each antibody and for each staining panel. With links to the authors Immunohistochemical Laboratory website, this volume creates a current and up-to-date information system on immunohistochemistry. This includes access to tissue microarrays (TMA) of over 5,000 tumors to validate common diagnostic panels and provide the best reproducible data for diagnostic purposes. Chapters are presented in a unique Question and Answer format. One table/IHC panel is provided to address each question. A concise explanatory note follows each table/panel to avoid diagnostic pitfalls. Website links are provided throughout to update the massive information in this field, providing the most current knowledge and the potential for live expert consultation. All chapters are written by nationally/internationally recognized experts in the related area ensuring authority and excellence. Comprehensive yet practical and concise, the Handbook of Practical Immunohistochemistry: Frequently Asked Questions, will be of great value for surgical pathologists, pathology residents and fellows, cytopathologists, and cytotechnologists.

This book examines a collection of state-of-the-art methods that employ monoclonal antibodies in a clinical setting. The chapters offer in-depth description for generating mouse and recombinant humanized antibodies, and a comprehensive review of how antibodies are being used in bead-based methods for measuring proteins. This field will continue to expand and provide new and innovative techniques in the laboratory and as a basis that complements targeted therapy.

Includes all of the information required to produce monoclonal antibodies in the laboratory and to prepare them for use in a multitude of given applications. Production procedures are treated in chronological order, beginning with basic tissue culture techniques, immunization strategies and screening test design, followed by production of hybridoma cell lines and basic antibody characterization, purification and labeling. Each chapter contains explanatory text on each step with comparative analysis of methods where appropriate. All necessary experimental protocols are presented in a self-contained format that is easy to follow in the laboratory. Alternative protocols are provided where relevant; for others not included in full, source references are presented. Surveys the current status of human hybridoma production and antibody engineering using molecular biology techniques.

FROM THE PREFACE: Several papers in this volume deal with advances in hybridoma technology and complement the chapter by Galfri and Milstein that appeared in Volume 73 of this series. Methods to enhance the efficiency of producing desired fusion products, to simplify the basic procedures involved, and to assess the specificity and other properties of monoclonal antibodies are discussed. Representative examples illustrate how a battery of monoclonal antibodies with different specificities can be used to study various immunochemical and biochemical problems. Other chapters cover additional procedures that can be used to label antigens and antibodies, separate antigen-antibody complexes, and process data. Some papers classified under one heading or contained in another volume of the series include methods that can be applied in other specific areas. For example, cytotoxicity tests described in Volume 93 may be used to screen supernatant fluids in hybridoma experiments if cytotoxic activity is a desirable property of the monoclonal antibody.

This volume provides a complete description of the principles, methodologies and applications of monoclonal antibodies, one of the most exciting developments to occur in biotechnology in recent years, and a powerful technology for modern industry and science. The immune system and the role of the antibody are described and full details are given on how the hybridomas are formed, isolated, and maintained in culture such that the required antibody can be produced to a high degree of purity. The authors describe all the methodologies involved, all the reagents and solutions and all the assay conditions required for their production. The material is presented to enable research and development managers to make choices as to which are the most suitable techniques for their requirements. The book is comprehensively referenced, and will be a necessary resource for all those who are involved with this technology.

This econometric study covers the world outlook for Monoclonal Antibody (MAb)-based products across more than 200 countries. For each year reported, estimates are given for the latent demand, or potential industry earnings (P.I.E.), for the country in question (in millions of U.S. dollars), the percent share the country is of the region and of the globe. These comparative benchmarks allow the reader to quickly gauge a country vis-a-vis others. Using econometric models which project fundamental economic dynamics within each country and across countries, latent demand estimates are created. This report does not discuss the specific players in the market serving the latent demand, nor specific details at the product level. The study also does not consider short-term cyclicalities that might affect realized sales. The study, therefore, is strategic in nature, taking an aggregate and long-run view, irrespective of the players or products involved.
This study does not report actual sales data (which are simply unavailable, in a comparable or consistent manner in virtually all of the 230 countries of the world). This study gives, however, my estimates for the worldwide latent demand, or the P.I.E. for Monoclonal Antibody (MAb)-based products. It also shows how the P.I.E. is divided across the world's regional and national markets. For each country, I also show my estimates of how the P.I.E. grows over time (positive or negative growth). In order to make these estimates, a multi-stage methodology was employed that is often taught in courses on international strategic planning at graduate schools of business.
