In the field of medicinal biotechnology three major developments have caused a revolution in research that has a lot of innovative effects on clinical medicine and future applications on humans. With the availability of tailored recombinant proteins and the opportunity to produce high amounts of monoclonal antibodies new diagnostic applications have emerged and many therapeutic perspectives, e.g. in the treatment of multiple sclerosis and of cancer, are being discussed today. The aim of somatic gene therapy is to re-establish normal cell function by supplying the cells with the respective intact gene. This is a very difficult task and different diseases, e.g. AIDS and several metabolic disorders, are under investigation now. So far first promising approaches exist in cancer therapy. Moreover the book informs about regulatory and economic aspects of these new methods and their applications.

Given the immense progress achieved in elucidating protein protein complex structures and in the field of protein interaction modeling, there is great demand for a book that gives interested researchers/students a comprehensive overview of the field. This book does just that. It focuses on what can be learned about protein protein interactions from the analysis of protein protein complex structures and interfaces. What are the driving forces for protein protein association? How can we extract the mechanism of specific recognition from studying protein protein interfaces? How can this knowledge be used to predict and design protein protein interactions (interaction regions and complex structures)? What methods are currently employed to design protein protein interactions, and how can we influence protein protein interactions by mutagenesis and small-molecule drugs or peptide mimetics? The book consists of about 15 review chapters, written by experts, on the characterization of protein protein interfaces, structure determination of protein complexes (by NMR and X-ray), theory of protein protein binding, dynamics of protein interfaces, bioinformatics methods to predict interaction regions, and prediction of protein protein complex structures (docking and homology modeling of complexes, etc.) and design of protein protein interactions. It serves as a bridge between studying/analyzing protein protein complex structures (interfaces), predicting interactions, and influencing/designing interactions.

Proteins are an integral part of molecular and cellular structure and function and are probably the most purified type of biological molecule. In order to elucidate the structure and function of any protein it is first necessary to purify it. Protein purification techniques have evolved over the past ten years with improvements in equipment control, automation, and separation materials, and the introduction of new techniques such as affinity membranes and expanded beds. These developments have reduced the workload involved in protein purification, but there is still a need to consider how unit operations linked together to form a purification strategy, which can be scaled up if necessary. The two Practical Approach books on protein purification have therefore been thoroughly updated and rewritten where necessary. The core of both books is the provision of detailed practical guidelines aimed particularly at laboratory scale purification. Information on scale-up considerations is given where appropriate. The books are not comprehensive but do cover the major laboratory techniques and common sources of protein. Protein Purification Techniques focuses on unit operations and analytical techniques. It starts with an overview of purification strategy and then covers initial extraction and clarification techniques. The rest of the book concentrates on different purification methods with the emphasis being on chromatography. The final chapter considers general scale-up considerations. Protein Purification Applications describes purification strategies from common sources: mammalian cell culture, microbial cell culture, milk, animal tissue, and plant tissue. It also includes chapters on purification of inclusion bodies, fusion proteins, and purification for crystallography. A purification strategy that can produce a highly pure single protein from a crude mixture of proteins, carbohydrates, lipids, and cell debris to is a work of art to be admired. These books (available individually or as a set)are designed to give the laboratory worker the information needed to undertake the challenge of designing such a strategy.

A complete account of the theory of the diffraction of x-rays by crystals with particular reference to the processes of determining the structures of protein molecules, this book is aimed primarily at structural biologists and biochemists but will also be valuable to those entering the field with a background in physical sciences or chemistry. It may be used at any post-school level, and develops from first principles all relevant mathematics, diffraction and wave theory, assuming no mathematical knowledge beyond integral calculus.
The book covers a host of important topics in the area, including: - The practical aspects of sample preparation and x-ray data collection, using both laboratory and synchrotron sources - Data analysis at both theoretical and practical levels - The important role played by the Patterson function in structure analysis by both molecular replacement and experimental phasing approaches - Methods for improving the resulting electron density map - The theoretical basis of methods used in refinement of protein crystal structures - In-depth explanation of the crucial task of defining the binding sites of ligands and drug molecules - The complementary roles of other diffraction methods which reveal further detail of great functional importance in a crystal structure.
