This practical volume gives real-life examples of how separation techniques are used in industry with recombinant proteins and emphasizes which techniques might be more useful than others. An introductory chapter ties together the various topics and associates the methods used with examples of large-scale methodologies. The emphasis of the volume is on how series can be applied to developing strategies and appropriate approaches for difficult protein separation problems. This valuable book provides an update on some of the existing technology used in industry and presents separation examples of several commercially important and recently developed proteins.

Proteins are an integral part of molecular and cellular structure and function and are probably the most purified type of biological molecule. In order to elucidate the structure and function of any protein it is first necessary to purify it. Protein purification techniques have evolved over the past ten years with improvements in equipment control, automation, and separation materials, and the introduction of new techniques such as affinity membranes and expanded beds. These developments have reduced the workload involved in protein purification, but there is still a need to consider how unit operations linked together to form a purification strategy, which can be scaled up if necessary. The two Practical Approach books on protein purification have therefore been thoroughly updated and rewritten where necessary. The core of both books is the provision of detailed practical guidelines aimed particularly at laboratory scale purification. Information on scale-up considerations is given where appropriate. The books are not comprehensive but do cover the major laboratory techniques and common sources of protein. Protein Purification Techniques focuses on unit operations and analytical techniques. It starts with an overview of purification strategy and then covers initial extraction and clarification techniques. The rest of the book concentrates on different purification methods with the emphasis being on chromatography. The final chapter considers general scale-up considerations. Protein Purification Applications describes purification strategies from common sources: mammalian cell culture, microbial cell culture, milk, animal tissue, and plant tissue. It also includes chapters on purification of inclusion bodies, fusion proteins, and purification for crystallography. A purification strategy that can produce a highly pure single protein from a crude mixture of proteins, carbohydrates, lipids, and cell debris to is a work of art to be admired. These books (available individually or as a set) are designed to give the laboratory worker the information needed to undertake the challenge of designing such a strategy.

A collection of key techniques for the study of receptors and transport proteins. The book provides examples of how different membrane proteins can be overexpressed in both prokaryotic and eukaryotic expression systems, how natural and overexpressed proteins can be solubilized from their host membranes, and how the solubilized protein can be purified in active form. Each protocols contains step-by-step instructions to ensure success, troubleshooting advice, lists of reagents, and tips on avoiding pitfalls.

The textbook is clearly written, and is suitable for advanced students, e.g. during their master and PhD course time. It is very suited also for postdocs and young scientists as a textbook if they change fields in their research. The textbook represents a thorough and detailed description of the topics in each of the chapters. As the book selects very important topics of present fundamental research and relevant in medical biochemistry, it can supplement any of the text books in biochemistry and basic medicine already on the market.

The second edition of "Structure in Protein Chemistry" showcases the latest developments and innovations in the field of protein structure analysis and prediction. The book begins by explaining how proteins are purified and describes methods for elucidating their sequences of amino acids and defining their posttranslational modifications. Comprehensive explanations of crystallography and of noncovalent forces - ionic interactions, hydrogen bonding, and the hydrophobic effect - act as a prelude to an exhaustive description of the atomic details of the structures of proteins. The resulting understanding of protein molecular structure forms the basis for discussions of the evolution of proteins, the symmetry of the oligomeric associations that produce them, and the chemical, mathematical, and physical basis of the techniques used to study their structures.

Upon publication of the first edition of Therapeutic Peptides and Proteins ten years ago there were only 19 biotechology medicines on the market. Currently there are more than 100, with at least 400 more in various stages of development. That alone would be grounds for a new edition. Add to that the fact that it is still difficult to find up-to-date, relevant information on protein formulation and/or delivery systems and the need for a new edition is self-evident. This second edition continues the tradition of providing a ready-to-use reference of accurate information, tables, and illustrations.

The book begins with the basics of the origin of recombinant proteins, explores structural and analytical aspects, and moves on to discuss the stability of proteins and peptides. Against this background, the author addresses preformulation and formulation considerations and provides detailed information on important topics such as lyophilization. He goes on to discuss various delivery systems, including controlled release delivery systems and products currently on the market. The book also contains newly added regulatory information and thoroughly investigates therapeutic drugs made from proteins as well as protein-based vaccines. A new chapter on recombinant protein subunit vaccines and their delivery systems has been added.

The work of a single author, the book provides complex information in a simple, elegant, and cohesive style. The author minimizes overlap between coverage and includes extensive cross-referencing that links related topics. Completely revised and updated, packed with cutting-edge information, the second edition once again fills the need for a reference that pulls all the relevant information together into one resource.
