In this, the post-genomic age, our knowledge of biological systems continues to expand and progress. As the research becomes more focused, so too does the data. Genomic research progresses to proteomics and brings us to a deeper understanding of the behavior and function of protein clusters. And now proteomics gives way to neuroproteomics as we begin to unravel the complex mysteries of neurological diseases that less than a generation ago seemed opaque to our inquiries, if not altogether intractable.

Edited by Dr. Oscar Alzate, Neuroproteomics is the newest volume in the CRC Press Frontiers of Neuroscience Series. With an extensive background in mathematics and physics, Dr. Alzate exemplifies the newest generation of biological systems researchers. He organizes research and data contributed from all across the world to present an overview of neuroproteomics that is practical and progressive.

Bolstered by each new discovery, researchers employing multiple methods of inquiry gain a deeper understanding of the key biological problems related to brain function, brain structure, and the complexity of the nervous system. This in turn is leading to new understanding about diseases of neurological deficit such as Parkinson’s and Alzheimer’s.

Approaches discussed in the book include mass spectrometry, electrophoresis, chromatography, surface plasmon resonance, protein arrays, immunoblotting, computational proteomics, and molecular imaging. Writing about their own work, leading researchers detail the principles, approaches, and difficulties of the various techniques, demonstrating the questions that neuroproteomics can answer and those it raises.

New challenges wait, not the least of which is the identification of potential methods to regulate the structures and functions of key protein interaction networks. Ultimately, those building on the foundation presented here will advance our understanding of the brain and show us ways to abate the suffering caused by neurological and mental diseases.

Description:
A novel protein was isolated from the seminal plasma of the mud crab, Scylla serrata (Forskal, 1775). It exhibited an antibacterial activity against the Gram-positive bacterium Micrococcus leteus with IC90 of 0.125 mg/ml. The extraction procedure for the protein included techniques of acid extraction, ion-exchange chromatography on SP-Sepharose Fast Flow and reverse-phase liquid chromatography on Source 5R RPC. It showed a molecular mass of 10.8 kDa by SDS-PAGE. A partial 20 residue NH"2-terminal sequence was determined by Edman degradation and MS-fingerprint of the protein was conducted. Similarity search in protein databases (BLAST) revealed that the protein exhibited no significant homology to any other reported antimicrobial peptides. We propose the name Scygonadin (from the gonad of S. serrata) for this antibacterial protein.

Exploring Proteins offers a complete course in developing the skills - and self-confidence - to be able to understand the way proteins behave and the basis of the methods used to separate, identify and characterise them.

Finding out about biological molecules and systems is a hands-on, experimental task: Exploring Proteins equips the reader with the skills they need to plan and carry out this experimental work, and to understand, analyse, and present the data their work generates.

With a focus throughout on active learning, the book blends background information on proteins and protein biochemistry, with the methods used to study them, featuring extensive worked examples and questions throughout to help the reader check their understanding, and develop critical data-handling skills for themselves.

It also offers complete flexibility - with sample data sets for readers without access to a lab, and suggested protocols for use where students can be given the opportunity to collect data for themselves.

This authoritative collection of basic and advanced protocols permits biotechnologists to use plants rather than single cell fermentation systems for expressing recombinant proteins. The protocols provide clear step-by-step instructions for the cloning, expression, and analysis of a wide variety of recombinant proteins in plants, and for the cloning of genes into a number of different plant species. The book also describes promising experimental techniques that will become increasingly important in the future, and includes techniques for the characterization of medically important proteins expressed in plants.

Paroxysmal Nocturnal Hemoglobinuria (PNH) has been recognized for over a century. This mysterious disease is now understood at the level of the gene and the protein. The pathophysiology is related to a class of cell surface proteins with distinctive biochemical and physical characteristics. Recently it has been acknowledged that PNH is not rare, and once sensitive assays-based on the chemistry of the proteins-can be applied to many patients. Written by international experts in the field, this book includes a number of distinctive characteristics, such as the clinical features of PNH, the mechanism of hemolysis, the biochemistry of glycosylphosphoinositol anchors, and the chemistry and biophysics of GPI-anchored proteins. This unique and timely volume will have a wide audience, including hematologists and oncologists with a clinical interest in this disease, as well as basic biochemists, immunologists, and cell biologists studying this class of proteins.

Part of a mini-series on the synthesis and processing of proteins, this volume details the expression of cloned DNA and RNA templates in all of the major in vivo and in vitro systems. The in vivo systems covered are cultured mammalian cells, the yeasts Sacchoromyces cerevisiae and Pichia pastoris, baculovirus, Xenopus oocytes, and prokaryotic cells. It also covers cell-free systems of both eukaryotes and prokaryotes, including prokaryotic systems that offer coupled transcription-translation. The volume concludes with a chapter on monitoring protein expression.

Since the discovery of the gene for green fluorescent protein (GFP), derived from jellyfish, this protein that emits a green glow has initiated a revolution in molecular biosciences. With this tool, it is now possible to visualize nearly any protein of interest in any cell or tissue of any species. Since the publication of the first edition, there have been tremendously significant technological advances, including development of new mutant variants. Proteins are now available in yellow and blue, and Novel Fluorescent Proteins (NFPs) have expanded their utility in developing biosensors, biological markers, and other biological applications.

This updated, expanded new edition places emphasis on the rise of NFPs, including new chapters on NFP properties with detailed protocols, applications of GFPs and NFPs in industry research, and biosensors. This book provides a solid theoretical framework, along with detailed, practical guidance on use of GFPs and NFPs with discussion of potential pitfalls. The expert contributors provide real examples in showing how to tailor GFP/NFP to specific systems, maximize expression, and enhance detection.
