hi,
I try to optimze luciferase assay in MCF10A and HCC1143 cell lines. During the transfection, I used Fugene HD reagent. I performed transfection with 490ng pmir vector and 10 ng renilla into serum and antibiotic free medium. After 6 or 24 hours, I change the medium. After 24 or 48 hours, I measured luciferase level. However I obtained different firefly:Renilla luciferase ratio in each experiment. Would you give me any idea to obtain consistent results?
