What test to use?

 I am really having trouble with the following question. I am not sure how I should choose what test to use or even where to start. I know the procedures for the various tests, but not sure on how to apply  to the question as I dont know which test is used best in what situation. If someone could help me with to this answer it, it would be greatly appreciated. Thank you.
 
You have been working hard in an immunology research group to create “knock-out” mice with homozygous deletions in the following genes: 1) Rag 1 & Rag 2 (double mutant); and 2) CD8.  Somehow, your mutant strains have been mixed up and now you must perform tests to determine which strain is which.  For each of the two mutant strains listed, briefly describe:
 
The specific immunological tests you would perform to distinguish between them. Choose from ELISA, Western Blot, immunoaffinity chromatography, immunofluorescent microscopy, and FACS techniques. Be sure to explain what particular reagents you would use, the expected results, and how these results will help you make the correct determination.
