Fluorescence Microscopy Optimization and Troubleshooting

I played with the condenser on the microscope manually. Normally when you look at a sample with white light on the confocal you get a nice round yellow circle on your slide. I am now getting a black circle in the middle. What is this? Is this related to the condenser? Rating: n/a
