Protocol for Intracellular Cytokine Staining by FACS

Protocol written by Protocol prepared by D.K. Mitra, S.C. De Rosa, N. Watanabe from Dr. Stephen C. De Rosa's lab at Stanford University.

The basic steps for intracellular staining of cytokine in cell culture:

- Harvest and pellet cells.
- Stain cells with surface marker antibodies and wash.
- Cell fixation with formaldehyde.
- Permeabilize cells and stain with cytokine antibodies for intracellular cytokine staining.
- Run FACS analysis.
