I am trying to express my favorite gene cloned in pET vector in
E. Coli BL21. The N-terminal truncated protein (missing 13AAs) can be
expressed to high level after 1mM IPTG induction for 2 to 4 hours.
However the full length protein can not be expressed at all. After the
2.5 ml overnight culture (some times this culture grew fine) was put
into 50 ml LB, the bacteria grew very very slow as compared with the truncated clone. I guess that ... ...

Es muy interesante lo que dice, pero necesito que me ayude a encontrar bibliografia sobre purificacion de enzimas de restriccion en E. coli, debido a que estoy proximo a graduarme como Microbiologo, por lo tanto estoy haciendo una monografia sobre el tema. |

hi, i am expressing a therapeutic protein in E.coli.after induction it does not grow O.D600 ,not more than 0.7.i feel it is toxic for E.coli, is ther any way to express this protein in e.coli.comments or suggestions are welcome.Thank you, Rajagopal.k |
