Double restriction digestion of DNA involves simultaneously DNA double digestion using two restriction enzymes. It is important to chose right enzymes with compatible buffers so that both enzymes have high activity. Most vendors provide restriction digestion buffer chart that allow users to check compatibility of different restriction digestion buffer system. NEB provides an online tool NEB double digestion enzyme finder to help users find compatible enzymes for double digestion. If there are difficulties to find compatible enzymes for double or even triple digestion, isoschizomers can be used and NEB has a extended list of Isoschizomers
