One of the most frequent laments of using recombinant retroviruses is that the titer does not appear to be sufficient to infect the cells of interest at a high enough frequency to suit your purposes.
This is a troubleshooting guide using Phoenix lines. (Nolan lab, Stanford University).

Dear sir
i am working as grad. astt. in ULM school of Pharmacy in the Pharmacoogy deptt. for a doctoral program. i am involved wih transfection procedures in TSA cell line using the CalPo4 system. i amnot able to get good transfection coz the scintillation count is not significant. can u guide me towards conducting stable transfection. i don't get solid lumps of DNA ppt. upon adding cal soln into the HEBS soln instead i get a very fine ppt. highly disperesed and the sytem appears light milky. Kindly guide me... Soliciting cooperation
