Glycerol stock longevity
Does any one have information (data or opinion or personal experience) on
how
long a bacterial glycerol stock is viable @ -80oC? How often to stocks fail
to
regrow? Is is necessary to thaw and revitalize the stock at time intervals?

I suspect that you did not elute the the 146bp band from the gel after the PCR reaction, and directly used the PCR cocktail or PCR purified product for A-tailing and/or ligation. You must be having some 800bp amplified fragment in your PCR mix which was not removed. so elute fragment from the gel. Rating: Very Good

My experiment have a puzzled trouble.Can any one kindly help me.
My target gene is 146bp in length.But when I got two colone(from total about 80 colones using promega easy T-vector),it seems that the colone contain 800bp insert.what caused that matter.could you tell me.pls.thank you Rating: Good
