mRNA purification protocol

mRNA purification protocol from Breeden's lab. The mRNA purification steps include preparing oligo-dT cellulose column, total RNA elution and washing in the oligo-dT cellulose column, precipitation and recovering of mRNA. Buffer recipes for mRNA purification are also included. (Breeden lab, Fred Hutchinson Cancer Research Center)

I Use RNeasy Kit (Qiagen cat. 74104) to clean up my total RNA before TaqmAn real-time PCR, or you can extract RNA directly with the Kit.
The Principle :
RNeasy silica-gel membrane allows up to 100 ug of RNA longer than 200 bp to bind , after wash, high quality RNA is eluted in water. With the RNeasy procedure, mRNA quality is enriched. Most of RNA< 200 bp such as rRNA, tRNA about 15 % of total RNA are excluded. see qiagenRNeasy hand book.
It's a very good kit so far I use for my RNA purification.
