TCA protein precipitation protocol

I would like to get some suggestions for better extraction pf proteins from calli of saffron. I have some problems in its IPG.

I carried out beta casein isolation from skim milk, by using ammonium sulfate.

however, after injecting my sample with the RE-HPLC the peake appear to be very small comparing with the puire standard of beta casein peak.

So I like to know how could I have I puire isolated beta casein by using Ammonium sulafte, and what kind of techniqueis I could use ferther to purified my sample

I usually use 12%trichloroacetic acid to precipitate meat protein samples. I know that in the supernatant remains amino acids and some little peptides. I need to know which is the size (approximately) of these peptides.In other words, I would to know the TCA cutt off for peptides if it is used as protein precipitant. Thank you in advance!

If I don't use the acetone purification step, (after thev TCA precipitation)can I have problemms? I have without acetone precipitation 5-6 bands of small proteins.

Hi,
I carried out a TCA precipitation and did get a white pellet, however, when i did a SDS-PAGE gel, I did not get any visible bands.
Did i do something wrong?
