we are working with the purification of the enzyme alkaline protease the crude enzyme shows good activity but once it is precipitated with ammonium sulfate and dialysed with a buffer tris of 50mM conc. it shows a very little activity by couplet method with casein. plz suggest me how do i improve the activity of my specific enzyme. thanku |

I have dialysed 5ml of my protein sample for the last 3 days with about 15 changes of 400 ml each in a dialysis bag of 10000 MW cut off. But the outside water is still showing a higher conductivity( 7.32 microsiemens) than pure water (4.3 microsiemens). Does it mean that it still has some salt? |
