I want to construct a DNA library with different sequence. First step,
PCR, the template contained random 40mers in the middle and normal
sequence on each end. After the PCR, I could obtain the PCR product

i am trying to clone a bacillus cellulases gene into pUC19.i cant get positive clones.what could be wrong?

i am using phage M13 to transfect Ecoli JM101 .but i am not getting blue colonies.what could be the problem
