Suspension Culture Infections: Low MOI
Infection of suspension cultures is the most common way of producing protein in the baculovirus system. Suspension cultures are easily scaleable from about 10 mL to more that 100 L. Generally, the suspension cultures are maintained in serum free media. These media are available from several commercial sources, all of which work well, though there is variation in effectiveness dependent upon cell line and protein. (Dr.Jim Litts, baculovirus.com)

Did you standardized you cell concentration in cultures? Difference in concentrations masks a lot. When I recalculated expression at 1 single cell , I did not fined any difference. |

I have experienced that when I infect cells in adherent culture the expression of protein is higher than in suspension culture. But this is not feasible because I have to produce the protein in large quantity. Please I need help on this. please reply to my email ibhatiahk@yahoo.com. thanks a lot |
