i am doing Ip and i can not get specific bands, but i get non specific bands |

The immunoprecipitation troubleshooting is of great help for troubleshooting common IP problems, such as non-specific background, specific background, no specific band detected, multiple specific bands detected etc. But how about two way or two directional immunoprecipitation problem? For example using A antibody can detect A-B complex but not with B antibody to detect A-B complex. Any suggestions for the possible cause? |

Immunoprecipitation(IP) is a very sensitive assay. The condirion of IP may be vary from different cells or tissues. General troubleshooting such as imcreasing preclearing to eliminate nonspecific bands may provide limited help. The only way to solve the problem is optimizing IP conditions through repeated trial with different factor adjustment. |
