hai, your streptavidin bead conc seems ok.how long is the spacer arm in your biotin . keep it as long as possible so that there would be minimum steric hinderance in later binding. i think this is critical. i use biotin- 21- dUTP which has 21-atom spacer arm, for 3'-end labelling and incorporate it through PCR. it worked fine in my clone capture procedure. |
