YAC DNA preparation

This protocol is used for YAC DNA extraction. YAC clones are provided as stab cultures in YPD agar. Glycerol stocks of each clone should be made as soon as possible. The clone will remain viable as a stab culture up to one week if stored at 4 c. (Hiroyuki Takeda lab in National Institute of Genetics, Japan)
