siRNA transfection protocols, technical considerations, resources and references are summarized. siRNA are 21-25 bp size double-stranded short RNA molecules that performs fundamental functions within cells and is becoming an important reverse genetics tool to study gene functions.

siRNA design is the critical step of a successful RNAi experiment. Well designed siRNA sequence will minimize the off-targeting effect and maximize the knock down effect of the given gene. Most commonly cited siRNA design tools and algorithms are collected here, along with some siRNA design tools from major RNAi suppliers.

One of the major problems of RNAi gene silencing in mammalian cells are non-specific effects related to off-target effects. The non-specific effect is triggered by partial complementarity of an RNAi when it is introduced into the cell.
